Multi-locus sequencing typing reveals geographic intraspecific variability of Sporothrix brasiliensis

Introdução

Sporotrichosis is a subcutaneous mycosis caused by Sporothrix species. This disease has been expanding throughout the Brazilian territory and its increasing number of cases has gained notoriety. Sporothrix brasiliensis is the main species isolated in Brazil. It is highly virulent and associated with zoonotic transmission. Moreover, S. brasiliensis has already been identified in Argentina and Paraguay, countries that borders Brazil. Molecular epidemiology survey is important to recognize genetic diversity, which could be related to outbreaks, and to identify different genotypes more or less susceptible to antifungal agents.

Objetivo(s)

In this study, our aim was establish a multi-locus sequence typing in order to identify variability among S. brasiliensis isolates

Material e Métodos

Analyses of four constitutive genes in 90 S. brasiliensis isolates from different Brazilian states from four regions of the country (South, Southeast, Midwest and Northeast) by DNA sequencing. These isolates were from clinical (n=26), veterinary (n=60), and environmental (n=2) sources. Besides, two S. brasiliensis isolates from Paraguay were also studied. Specific primers were designed for 15 genes using Primer-BLAST software from the S. brasiliensis whole genome available in the Genbank. The discriminatory and selection of the loci that presented more variability were made by DNAsp v5.0.

Resultados e Conclusão

The PCR was standardized for all genes and, among them, four loci were able to separate the S. brasiliensis isolates in six distinct haplotypes, which were related to their geographic distribution. Human isolates grouped with both veterinary and environmental isolates, reinforcing S. brasiliensis transmission from both classic and zoonotic forms. This study presents a great progress related to the epidemiology of the most important sporotrichosis agent in Brazil. Besides the knowledge about the genetic diversity of S. brasiliensis, our results add significant data about sporotrichosis in Brazil and in others countries.
